The regulation of enzymatic activity is a major event in nature as well as in different scientific research areas like medicinal chemistry, molecular biology or biotechnology. Besides the control with small molecules (activators, inhibitors) it is shown that enzymatic activity can also be regulated by irradiation with light. In this project, we generate a light switchable model approach with the T7 RNA Polymerase as enzymatic effector and the Light-Oxygen-Voltage (LOV) domain as photosensor to design a photocontrollable DNA Transcription system in vitro and in vivo.
